What is Daraprim?

Daraprim contains the prescription medicine pyrimethamine. Pyrimethamine is classed as an antiparasite medicine, used to stop parasites from growing and multiplying in the body.

Daraprim is used to treat certain types of malaria in adults and children. Daraprim is not the preferred choice of medicine to prevent malaria when traveling however, as there is substantial resistance to pyrimethamine worldwide. Daraprim should be used together with other faster-acting anti-malaria medicine such as chloroquine or quinine to treat malaria.

Toxoplasmosis, an infection caused by the Toxoplasma parasite is also treated using Daraprim, in combination with other medicines known as sulfonamides.

Daraprim active ingredients

Pyrimethamine, the active ingredient in Daraprim, blocks the reproduction of the parasite once it is in the hosts’ bloodstream. It does this by interfering with the production of a chemical called tetrahydrofolic acid from dihydrofolate. Tetrahydrofolic acid is vital for DNA production for the parasite. Without this, the parasite is unable to survive.

The effect of pyrimethamine against Toxoplasmosis is far greater when used in combination with sulfonamides.

Daraprim side effects

The most common side effects caused by Daraprim include:

  • Loss of appetite
  • Vomiting

In rare instances, Daraprim can cause more serious side effects. These can include:

  • Irregular heart rhythm
  • Blood in your urine
  • Lack of folate – feeling tired or sick, stomach upset, mood swings, mouth ulcers, loss of appetite
  • Swollen tongue, sore throat
  • Skin rash
  • Pale skin, easily bruised, purple spots under the skin
  • Cold or flu symptoms, cough, fever, difficulty breathing

Your doctor will assess the benefits of using Daraprim against your risk of side effects.

Daraprim drug interactions

Daraprim can interact with other medications. These include:

  • Methotrexate
  • Phenytoin or lamotrigine, used to treat seizures
  • Sulfa drugs such as sulfamethoxazole, sulfadiazine, or sulfasalazine
  • Aurothiglucose or auranofin used to treat rheumatoid arthritis
  • Deferiprone, used to remove excess iron after treatment for thalassemia
  • Gold sodium thiomalate

This list is not exhaustive and other drugs may interact with Daraprim.

Daraprim warning and precautions

You should not use Daraprim if you:

  • Are allergic to the active ingredient pyrimethamine
  • Are allergic to any of the other ingredients in Daraprim
  • Have megaloblastic anemia caused by a lack of folate

Talk to your doctor before using Daraprim if you:

  • Are taking any of the medications that could interact with Daraprim
  • Suffer from seizures, especially if you are taking phenytoin
  • Have liver disease
  • Have kidney disease
  • Suffer from alcoholism
  • If you are malnourished
  • Are pregnant or are planning to become pregnant
  • Are breastfeeding or are planning to breastfeed
