What is Daurismo?

Daurismo is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Pfizer Inc. Daurismo is a hedgehog pathway inhibitor indicated, in combination with low-dose cytarabine, for the treatment of newly-diagnosed acute myeloid leukemia (AML) in adult patients who are ≥75 years old or who have comorbidities that preclude the use of intensive induction chemotherapy. Daurismo is not recommended in patients with the comorbidities of severe renal impairment or moderate-to-severe hepatic impairment.

Warning – Embryo-fetal toxicity. See the full prescribing information for the complete boxed warning. Daurismo can cause embryo-fetal death or severe birth defects when given to a pregnant woman. Daurismo is embryotoxic, fetotoxic, and teratogenic in animals. Carry out pregnancy testing in females of reproductive potential prior to initiation of treatment. Advise females of reproductive potential to use effective contraception during treatment and for at least 30 days after the last dose. Advise males of the potential risk of exposure through semen and to use condoms with a pregnant partner or a female partner of reproductive potential during treatment and for at least 30 days after the last dose.

What doses of Daurismo are available?

Daurismo is available in tablet form, in the following doses: 25 mg, 100 mg.

Daurismo side effects

The most common adverse reactions of Daurismo in clinical trials include:

  • Anemia (decreased red blood cells)
  • Febrile neutropenia (decreased white blood cell count)
  • Thrombocytopenia (decreased platelet count)
  • Tiredness, hemorrhage
  • Musculoskeletal pain, muscle pain, muscle spasms
  • Nausea
  • Edema
  • Dyspnea
  • Decreased appetite
  • Dysgeusia
  • Mucositis
  • Constipation
  • Rash

In rare instances, Daurismo can cause more serious adverse events. These can include:

  • QTc interval prolongation – monitor electrocardiograms and electrolytes. Patients treated with Daurismo can develop QTc prolongation and ventricular arrhythmias, including ventricular fibrillation and ventricular tachycardia.

Daurismo drug interactions

Daurismo can interact with other medications. These include:

  • Strong CYP3A4 inhibitors
  • Strong CYP3A4 inducers
  • QTc prolonging drugs – avoid co-administration, if unavoidable, monitor for increased risk of QTc interval prolongation

This list is not exhaustive and other prescription drugs may interact with Daurismo.

Daurismo contraindications

You should not use Daurismo if you:

  • Are allergic to the active ingredient glasdegib
  • Are allergic to any of the other ingredients in Daurismo
  • Are pregnant or are planning to become pregnant
  • Are breastfeeding or are planning to breastfeed

Talk to your doctor before using Daurismo if you:

  • Are taking any of the medications that could interact with Daurismo

Daurismo generic

Daurismo is the trademark brand name for glasdegib manufactured by Pfizer Inc. A generic version of glasdegib is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Daurismo savings through NiceRx.
