What is Empaveli?

Empaveli is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Apellis Pharmaceuticals, Inc. It is a complement inhibitor indicated for the treatment of adult patients with paroxysmal nocturnal hemoglobinuria (PNH) who are:

  • Treatment naïve
  • Switching from C5 inhibitor Soliris (eculizumab)
  • Switching from C5 inhibitor Ultomiris (ravulizumab)

PNH is a rare, chronic blood disorder characterized by the breakdown of healthy, red blood cells (anemia) by an uncontrolled immune system, blood clots, and defective bone marrow not making enough blood components.

What doses of Empaveli are available?

Empaveli is available in injection form, in the following doses: 1,080 mg/20 mL in a single-dose vial.

The recommended dosage is 1,080 mg by subcutaneous infusion twice weekly.

Empaveli may predispose patient to serious infections, especially those caused by encapsulated bacteria, such as Streptococcus pneumoniae, Neisseria meningitidis types A, C, W, Y, and B, and Haemophilus influenzae type B. Patients should be vaccinated against encapsulated bacteria at least 2 weeks before the first dose of Empaveli unless the risks of delaying treatment outweigh the risks of developing a serious infection.

Empaveli is available only through the Empaveli REMS restricted program under a Risk Evaluation and Mitigation Strategy (REMS).

Empaveli active ingredients

The active ingredient of Empaveli is pegcetacoplan.

Empaveli side effects

The most common side effects of Empaveli in clinical trials include:

  • Injection-site reactions
  • Diarrhea, abdominal pain
  • Respiratory tract infection, viral infection
  • Tiredness

In rare instances, Empaveli can cause more serious side effects. These can include:

  • Meningococcal infections that may be life-threatening or fatal
  • Infusion-related Reactions
  • Interference with laboratory tests
  • Allergic reactions – chest pain, difficulty breathing or shortness of breath, swelling of your face, tongue, or throat, feeling light-headed

Empaveli drug interactions

Other drugs may interact with Empaveli, including prescription drugs and over-the-counter medicines, vitamins, and herbal products. Tell your health care provider about all of your medication including any supplements you are taking.

Empaveli contraindications

You should not use Empaveli if you:

  • Have hypersensitivity to the active ingredient pegcetacoplan
  • Have had an allergic reaction to any of the other ingredients in Empaveli
  • Are not currently vaccinated against certain encapsulated bacteria unless the risks of delaying treatment outweigh the risks of developing serious bacterial infection with an encapsulated organism
  • Are pregnant or are planning to become pregnant – use effective contraception during treatment and for 40 days after the last dose

Talk to your doctor before using Empaveli if you:

  • Are taking any of the medications that could interact with Empaveli
  • Are breastfeeding or are planning to breastfeed

Empaveli generic

Empaveli is the trademark brand name for pegcetacoplan manufactured by Apellis Pharmaceuticals, Inc. A generic version of pegcetacoplan is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Empaveli savings through NiceRx.
