What is Fyarro?

Fyarro is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Aadi Bioscience, Inc. Fyarro is an mTOR inhibitor bound to human albumin. FDA approval for Fyarro was based on data from the multicenter phase 2 AMPECT trial which evaluated the efficacy of Fyarro in adults aged 18 years and older with locally advanced malignant PEComa or metastatic malignant PEComa. Patients were given Fyarro intravenously weekly until disease progression or unacceptable toxicity. The primary endpoint was the overall response rate (ORR).

Fyarro is indicated for the treatment of adult patients with locally advanced unresectable or metastatic malignant perivascular epithelioid cell tumor (PEComa).

What doses of Fyarro are available?

Fyarro is available as a lyophilized powder containing 100 mg of sirolimus formulated as albumin-bound particles in a single-dose vial for reconstitution as an injectable suspension.

Fyarro side effects

The most common side effects of Fyarro in clinical trials include:

  • Stomatitis, fatigue, rash, infection, nausea, edema, diarrhea, musculoskeletal pain, weight loss, decreased appetite, cough, vomiting, and dysgeusia
  • Laboratory abnormalities include decreased lymphocytes, decreased potassium, decreased phosphate, decreased hemoglobin, increased lipase, and increased glucose

In rare instances, Fyarro can cause more serious adverse effects. These can include:

  • Interstitial lung disease (ILD)
  • Myelosuppression
  • Infections – may result from immunosuppression
  • Hypokalemia and hyperglycemia
  • Non-infectious pneumonitis
  • Hemorrhage
  • Hypersensitivity reactions
  • Embryo-Fetal Toxicity
  • Male infertility

Fyarro drug interactions

Fyarro can interact with other medications. These include:

  • Strong CYP3A4 and/or P-gp inhibitors or inducers
  • Moderate or weak CYP3A4 inhibitors
  • Immunizations -avoid live vaccines
  • Grapefruit juice or grapefruit

This list is not exhaustive and other prescription drugs may interact with Fyarro.

Fyarro contraindications

You should not use Fyarro if you:

  • Are allergic to the active ingredient sirolimus protein-bound particles for injectable suspension (albumin-bound)
  • Are allergic to any of the other ingredients in Fyarro
  • Have a history of severe hypersensitivity to sirolimus, other rapamycin derivatives, or albumin
  • Are pregnant or are planning to become pregnant

Talk to your doctor before using Fyarro if you:

  • Are taking any of the medications that could interact with Fyarro
  • Have hepatic Impairment
  • Are breastfeeding or are planning to breastfeed – Do not breastfeed during treatment and for 2 weeks after your last dose

Fyarro is the trademark brand name for sirolimus protein-bound particles for injectable suspension (albumin-bound) manufactured by Aadi Bioscience, Inc. A generic version of sirolimus protein-bound particles for injectable suspension (albumin-bound) is not available.
