What is Gamifant?

Gamifant is a medicine used in rare diseases, with FDA approval (U.S. Food and Drug Administration) for the treatment of HLH. It is manufactured by Novimmune and distributed by Sobi Inc.

Gamifant is an interferon-gamma (IFNγ) blocking antibody for the treatment of adult and pediatric (newborn and older) patients with primary hemophagocytic lymphohistiocytosis (HLH) with refractory, recurrent or progressive disease or intolerance with conventional HLH therapy.

Primary HLH is a genetic disease characterized by an overactive immune system, in which the body produces too many macrophages and lymphocytes. The macrophages and the lymphocytes should destroy infected and damaged cells of the body, but in HLH the immune system damages the patient’s healthy cells.

What doses of Gamifant are available?

Gamifant is available in injection form, in the following doses: 10 mg/2 mL (5 mg/mL) solution in a single-dose vial, 50 mg/10 mL (5 mg/mL) solution in a single-dose vial.

The recommended starting dosage is 1 mg/kg as an intravenous infusion, over 1 hour, twice per week. Administer dexamethasone at the same time with Gamifant.

Use Gamifant until hematopoietic stem cell transplantation (HSCT) a procedure where the patient’s bone marrow is replaced by stem cells from a donor to form new bone marrow that produces healthy cells, is performed or unacceptable toxicity.

Always speak with your healthcare provider about any changes to your dose so they can monitor and evaluate your condition.

Gamifant active ingredients

The active ingredient of Gamifant is Emapalumab-lzsg, a monoclonal antibody that binds to and neutralizes interferon-gamma (IFNγ).

The main measure of the effectiveness of Gamifant in clinical studies was the number of patients who had a partial response or complete response or HLH improvement, based on improvement in their symptoms and results of laboratory tests after 4 to 8 weeks of treatment.

Gamifant side effects

The most common side effects of Gamifant in clinical trials include:

  • Hypertension
  • Infusion-related reactions
  • Pyrexia

In rare instances, Gamifant can cause more serious side effects. These can include:

  • Infections – administer prophylactic treatment against Herpes zoster, pneumocystis jirovecii, and fungal infections
  • Infusion-related reactions
  • New or worsening signs of infection

Your caregiver will assess the benefits of using Gamifant against your risk of side effects. Patients are encouraged to report negative side effects or adverse reactions of Gamifant to the FDA. Visit www.fda.gov/medwatch, or call 1-800-FDA-1088.

Gamifant drug interactions

Gamifant can interact with other medications. These include:

  • The formation of CYP450 enzymes may be blocked by increased levels of cytokines (such as IFNγ) during chronic inflammation
  • Live vaccines

Gamifant contraindications

You should not use Gamifant if you:

  • Are allergic to the active ingredient emapalumab-lzsg
  • Are allergic to any of the other ingredients in Gamifant

Talk to your doctor before using Gamifant if you:

  • Are taking any of the medications that could interact with Gamifant
  • Are pregnant or are planning to become pregnant
  • Are breastfeeding or are planning to breastfeed
  • Have ever had tuberculosis or if anyone in your household has tuberculosis
  • Have recently traveled – tuberculosis and some fungal infections are more common in certain parts of the world, and you may have been exposed during travel
  • Have any type of infection (active or recent)
  • Have a fungal infection
  • Have herpes zoster
  • Are scheduled to receive a vaccine

Gamifant generic

Gamifant is the trademark brand name for emapalumab-lzsg manufactured by Novimmune SA, Geneva, Switzerland, U.S. License Number 2082. Distributed by: Sobi Inc. A generic version of emapalumab-lzsg is available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Gamifant savings through NiceRx.
