What is Ingrezza?

Ingrezza is an FDA (Food and Drug Administration) approved medication manufactured by Neurocrine Biosciences, Inc., San Diego. It contains the prescription medicine valbenazine. Ingrezza is used for the treatment of a condition known as tardive dyskinesia in adults. Tardive dyskinesia is a nervous disorder that can develop due to the long-term use of antipsychotic drugs. Symptoms are the uncontrolled repetitive movement of muscles (akathisia) in the face, tongue, or other parts of the body.

Ingrezza is not a cure for tardive dyskinesia and it has not been established if it is safe for use or effective in children.

Ingrezza should be taken as prescribed by your doctor, with or without food. You may need to have an ECG (electrocardiograph) to check your heart function while taking this medicine. You are advised not to breastfeed while taking Ingrezza or for at least 5 days after you have stopped. You should not stop taking this medication unless your healthcare provider advises you to do so.

What doses of Ingrezza are available?

Ingrezza is available in capsule form, in the following doses: 40 mg and 80 mg. Always speak with your healthcare provider about any changes to your dose so they can monitor and evaluate your condition.

Ingrezza active ingredients

Valbenazine is thought to work by blocking the action of what is known as vesicular monoamine transporter 2 (VMAT2). This transporter normally controls the movement of a chemical called monoamine from the cytoplasm in cells to synaptic vesicles for storage and release. Synaptic vesicles are involved in how electrical signals (nerve impulses) are sent through your body.

Clinical trials with Ingrezza show after 6 weeks of treatment, 40% of people taking the 80 mg capsule of Ingrezza daily had at least a 50% decrease in their symptoms. Compared to 8.7% of people taking a placebo had the same result.

The exact mechanism of action of valbenazine for the treatment of tardive dyskinesia is, however, not fully understood.

Ingrezza side effects

The most common side effects and adverse reactions caused by Ingrezza include:

  • Headache
  • Anticholinergic effects – blurred vision, dizziness, balance disorders, dry mouth
  • Increased risk of falls
  • Arthralgia – joint stiffness
  • Constipation

In rare instances, Ingrezza can cause more serious side effects. These can include:

  • Heart rhythm problems (QT prolongation)
  • Somnolence (sleepiness)
  • Shortness of breath
  • Feeling light-headed or faint

Your doctor will assess the benefits of using Ingrezza against your risk of side effects. You may report the side effects of prescription drugs to FDA at 1-800-FDA-1088.

Ingrezza drug interactions

Ingrezza can interact with other medications. These include:

  • Antidepressants known as SSRI (selective serotonin reuptake inhibitor) – paroxetine and fluoxetine
  • Medication used to treat irregular heartbeats – quinidine and digoxin
  • Antibiotics used for the treatment of bacterial infections – rifampin
  • Medication that is taken to prevent seizures – carbamazepine and phenytoin
  • St. John’s wort, a herbal medication used for depression and mood disorders
  • Strong CYP3A4 such as itraconazole, ketoconazole, or clarithromycin
  • Monoamine oxidase inhibitors (MAOIs) – isocarboxazid, phenelzine, selegiline

This list is not exhaustive and other prescription drugs may interact with Ingrezza. Also, tell your doctor about any vitamins, herbs, and supplements you use.

Ingrezza contraindications

You should not use Ingrezza if you:

  • Are allergic to the active ingredient valbenazine
  • Are allergic to any of the other ingredients in Ingrezza
  • Are younger than 18 years old

Talk to your doctor before using Ingrezza if you:

  • Are taking any of the medications that could interact with Ingrezza
  • Have or suffer from any form of heart disease
  • Have a condition known as long QT syndrome or any other heart rhythm disorder
  • Have congestive heart failure
  • Have previously had a heart attack
  • Have liver disease
  • Have kidney disease
  • Are breastfeeding or are planning to breastfeed
  • Are pregnant or are planning to become pregnant

You should not breastfeed while using this medicine and for at least 5 days after you stop.

Ingrezza generic

Ingrezza is the trademark brand name for valbenazine, manufactured by Neurocrine Biosciences, Inc. A generic version of Ingrezza is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Ingrezza savings through NiceRx.
