What is Palforzia?

Palforzia [Peanut (Arachis hypogaea) Allergen Powder-dnfp] is an FDA-approved (Food and Drug Administration) medication manufactured by Aimmune Therapeutics, Inc.

Palforzia is oral immunotherapy for the treatment of allergic reactions, including anaphylaxis, that may occur with accidental exposure to peanuts.

Palforzia is approved for use in patients with a confirmed diagnosis of peanut allergy. Peanut allergies are one of the most common food allergies in the world, affecting more than 1.6 million children and teens in the United States alone.

Initial dose escalation may be given to patients aged 4 to 17 years of age. Up-dosing and maintenance treatment may be continued in patients 4 years of age and older. Palforzia is to be used in combination with a peanut-avoidant diet.

It is only available through the Palforzia Risk Evaluation and Mitigation Strategy (REMS) program with a certified healthcare provider.

What doses of Palforzia are available?

Palforzia is in capsule or powder for oral administration form, in the following doses: 0.5 mg 1 mg, 10 mg, 20 mg, and 100 mg Capsules or 300 mg Sachets.

You will receive your first dose of Palforzia treatment or any increases in dose in a hospital or healthcare setting by a healthcare professional. You will be monitored closely after being given your first dose.

Do not swallow a Palforzia capsule whole or inhale the powder medicine. Open the capsule or powder and mix the medicine into a spoonful of applesauce, yogurt, or pudding (do not mix with liquid).

You must keep injectable epinephrine with you at all times while you are using Palforzia. Read and carefully follow any Instructions for use provided with your epinephrine.

Take Palforzia as prescribed by your doctor. Follow all directions on your prescription label and medication guides. Your doctor may occasionally change your dosing levels. Always speak with your allergist about any changes to your dose so they can monitor and evaluate your condition.

Palforzia active ingredients

Palforzia contains peanuts (Arachis hypogaea) which gives children with a peanut allergy controlled exposure to precise amounts of peanut protein every day. This may help them decrease their sensitivity to small amounts of peanuts over time.

Palforzia side effects

The most common side effects of Palforzia in clinical trials include:

  • Abdominal pain vomiting, nausea
  • Oral pruritus, oral paresthesia – itching or burning in your mouth
  • Throat irritation, cough, rhinorrhea, sneezing, throat tightness
  • Wheezing, dyspnea, pruritus, urticaria, anaphylactic reaction
  • Ear pruritus – itching around the ear

In rare instances, Palforzia can cause more serious side effects. These can include:

  • Hives, chest discomfort, wheezing, tightness in your throat, trouble speaking or swallowing, swelling of your face, lips, eyes, or tongue, sudden warmth, redness, or tingly feeling, severe stomach pain, vomiting, or diarrhea, light-headed feeling, like you might pass out
  • Your caregiver may prescribe epinephrine injection in case you have a risk of anaphylaxis, (a severe allergic reaction). Get emergency medical help if you have an allergic reaction, even if you have used epinephrine to treat it

Palforzia drug interactions

Palforzia contraindications

You should not use Palforzia if you:

  • Are allergic to the active ingredient Peanut (Arachis hypogaea)

Talk to your doctor before using Palforzia if you:

  • Are pregnant or are planning to become pregnant
