What is Pemazyre?

Pemazyre is an FDA (U.S. Food and Drug Administration) approved medication developed by Incyte Corporation. It is a kinase inhibitor for the treatment of adults with previously treated, unresectable locally advanced or metastatic cholangiocarcinoma (bile duct cancer) with a fibroblast growth factor receptor 2 (FGFR2) fusion or other rearrangements.

This indication is approved under accelerated approval based on the overall response rate (ORR) and duration of response (DOR). Continued approval for this indication may depend upon verification and description of clinical benefit in a confirmatory trial(s).

What doses of Pemazyre are available?

Pemazyre is available in tablet form, in the following doses: 4.5 mg, 9 mg, and 13.5 mg.

Before starting treatment confirm the presence of an FGFR2 fusion or rearrangement. The recommended dose is 13.5 mg orally once daily for 14 days followed by 7 days off therapy in 21-day cycles. Continue treatment until disease progression or unacceptable toxicity occurs. Swallow the tablets whole, with or without food.

Pemazyre active ingredients

The active ingredient of Pemazyre is pemigatinib.

Pemazyre side effects

The most common adverse reactions (incidence ≥ 20%) of Pemazyre in clinical trials include:

  • Alopecia (hair loss)
  • Diarrhea, constipation
  • Nail toxicity
  • Fatigue
  • Dysgeusia, stomatitis, dry mouth, nausea,
  • Dry eye, ocular toxicity
  • Nausea, vomiting, abdominal pain
  • Arthralgia (joint pain), back pain
  • Hypophosphatemia
  • Dry skin

In rare instances, Pemazyre can cause more serious side effects. These can include:

  • Retinal pigment epithelial detachment
  • Hyperphosphatemia – increases in phosphate levels

Pemazyre drug interactions

Pemazyre can interact with other medications. These include:

  • Strong and moderate CYP3A Inducers – avoid concomitant use of Pemazyre
  • Strong and moderate- CYP3A inhibitors

Pemazyre contraindications

You should not use Pemazyre if you:

  • Are allergic to the active ingredient pemigatinib
  • Are allergic to any of the other ingredients in Pemazyre
  • Are pregnant or are planning to become pregnant – a potential risk to the fetus of pregnant women. Female patients of reproductive potential are advised to use effective contraception during treatment and for 1 week after the final dose
  • Are breastfeeding or are planning to breastfeed

Talk to your doctor before using Pemazyre if you:

  • Are taking any of the medications that could interact with Pemazyre
  • Have a severe hepatic impairment
  • Have a severe renal impairment

Pemazyre generic

Pemazyre is the trademark brand name for pemigatinib developed by Incyte Corporation. A generic version of pemigatinib is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Pemazyre savings through NiceRx.
