What is Qinlock?

Qinlock is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Deciphera Pharmaceuticals, Inc. Qinlock is a tyrosine kinase inhibitor indicated for the treatment of adult patients with advanced gastrointestinal stromal tumor (GIST) who have received prior treatment with 3 or more kinase inhibitor therapies including imatinib.

FDA approval of Qinlock was based on the Invictus study, a phase three randomized, international, multi-center, double-blind, placebo-controlled clinical trial. Patients were those with advanced GIST who had received prior treatment with other targeted therapies, imatinib, sunitinib, and regorafenib.

The trial compared patients who were randomized to receive Qinlock against those randomized to receive placebo, to determine whether progression-free survival (PFS) was longer in the Qinlock group or the placebo group. Patients received Qinlock or placebo until disease progression, or the patient experienced intolerable side effects. On average, the PFS rate in patients in the Qinlock group was 6.3 months, compared to one month for patients in the placebo group.

Most cases of GIST are caused by a variety of mutations. Proto-oncogene c-KIT kinase accounts for 80% of cases, platelet-derived growth factor receptor α (PDGFRA) kinase accounts for 10% of cases.

What doses of Qinlock are available?

Qinlock is available in tablet form, in the following doses: 50 mg.

The recommended dosage is 150 mg orally once daily with or without food.

Qinlock side effects

The most common side effects of Qinlock in clinical trials compared to placebo include:

  • Alopecia (hair loss)
  • Tiredness
  • Nausea, vomiting
  • Abdominal pain
  • Constipation, diarrhea, decreased appetite
  • Myalgia (muscle pain)

In rare instances, Qinlock can cause more serious side effects. These can include:

  • Palmar-plantar erythrodysesthesia syndrome
  • New primary cutaneous malignancies
  • Hypertension
  • Cardiac dysfunction – assess ejection fraction by echocardiogram or MUGA scan before starting and during treatment
  • Impaired wound healing
  • Embryo-fetal toxicity

Qinlock contraindications

You should not use Qinlock if you:

  • Are allergic to the active ingredient ripretinib
  • Are allergic to any of the other ingredients in Qinlock
  • Are pregnant or are planning to become pregnant
  • Are breastfeeding or are planning to breastfeed

Talk to your doctor before using Qinlock if you:

  • Are taking any of the medications that could interact with Qinlock

Qinlock is the trademark brand name for ripretinib manufactured by Deciphera Pharmaceuticals, Inc. A generic version of ripretinib is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Qinlock savings through NiceRx.
