What is Revcovi?

Revcovi is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Leadiant Biosciences Inc. It is an enzyme replacement therapy (ERT) used to treat adenosine deaminase deficiency (ADA-SCID) in pediatric and adult patients. ADA-SCID with a deficiency of ADA enzyme is classed as a rare disease and is a genetic disorder. Revcovi helps increase the number of blood cells (lymphocytes) that prevent and fight infection.

What doses of Revcovi are available?

Revcovi is available as an intramuscular injection, in the following doses: 2.4 mg/1.5 mL in a single-dose vial.

For patients moving from Adagen to Revcovi, the starting dose is 0.2 mg/kg weekly, intramuscularly. For Adagen-naïve patients, the starting dose of is 0.4 mg/kg weekly based on ideal body weight.

Once treatment has started, a target trough plasma ADA activity should be at least 30 mmol/hr/L. Decreased activity, below 30 mmol/hr/L suggests noncompliance to treatment or antibody development. 2 months after starting Revcovi treatment, trough erythrocyte dAXP levels should be maintained below 0.02 mmol/L.

Revcovi active ingredients

Revcovi contains elapegademase-lvlr, a recombinant adenosine deaminase, as the active ingredient. Elapegademase-lvlr provides an external source of ADA enzyme that is linked to a decrease in toxic adenosine and deoxyadenosine nucleotides levels as well as an increase in lymphocytes

Revcovi side effects

The most common side effects of Revcovi in clinical trials include:

  • Cough
  • Vomiting

In rare instances, Revcovi can cause more serious side effects. These can include:

  • Risk of infection
  • Delay in improvement of immune function. Protect patients with immune system deficiency from infections until improvement in immune function
  • Injection site bleeding in patients with thrombocytopenia

Revcovi drug interactions

Other drugs may interact with Revcovi, including prescription drugs and over-the-counter medicines, vitamins, and herbal products. Tell your health care provider about all of your medications including any supplements you are taking.

Revcovi contraindications

You should not use Revcovi if you:

  • Are allergic to the active ingredient elapegademase-lvlr
  • Are allergic to any of the other ingredients in Revcovi

Talk to your doctor before using Revcovi if you:

  • Are taking any of the medications that could interact with Revcovi
  • Are pregnant or are planning to become pregnant
  • Are breastfeeding or are planning to breastfeed

Revcovi is the trademark brand name for elapegademase-lvlr manufactured by Leadiant Biosciences Inc. A generic version of elapegademase-lvlr is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Revcovi savings through NiceRx.
