What is Rybrevant?

Rybrevant is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Janssen Pharmaceutical Companies of Johnson & Johnson. It is a bispecific EGF receptor-directed and MET receptor-directed antibody.

Rybrevant is used for the treatment of adult patients with locally advanced or metastatic non-small cell lung cancer (NSCLC) with epidermal growth factor receptor, EGFR exon 20 insertion mutations, whose disease has progressed on or after platinum-based chemotherapy.

This indication is approved under accelerated approval based on overall response rate (ORR) and duration of response (DOR). Continued approval for this indication may depend upon verification and description of clinical benefit in the confirmatory trials.

What doses of Rybrevant are available?

Rybrevant is available in injection form, in the following doses: 350 mg/7 mL (50 mg/mL) solution in a single-dose vial.

Premedications should be administered as recommended. Rybrevant should be given until disease progression or unacceptable toxicity. Please read the full prescribing information.

Rybrevant active ingredients

The active ingredient of Rybrevant is amivantamab-vmjw, a bispecific antibody that binds to the extracellular domains of EGFR and MET on the surface of tumor cells, allowing for the targeting of these cells.

Rybrevant side effects

The most common side effects of Rybrevant in clinical trials include:

  • Dry skin
  • Paronychia
  • Musculoskeletal pain
  • Dyspnea, stomatitis, cough
  • Nausea, vomiting
  • Fatigue, shortness of breath
  • Edema
  • Dry eye
  • Constipation
  • Blood tests show decreased lymphocytes, albumin, phosphate, potassium, and sodium, increased alkaline phosphatase, glucose, gamma-glutamyl transferase

In rare instances, Rybrevant can cause more serious adverse reactions. These can include:

  • Infusion-Related Reactions (IRR)
  • Interstitial Lung Disease (ILD)/Pneumonitis
  • Dermatologic Adverse Reactions
  • Ocular Toxicity

Rybrevant drug interactions

Rybrevant contraindications

You should not use Rybrevant if you:

  • Are allergic to the active ingredient amivantamab-vmjw
  • Are allergic to any of the other ingredients in Rybrevant
  • Are pregnant or are planning to become pregnant – can cause embryo-fetal toxicity in pregnant women. Females of reproductive potential are advised to use effective contraception
  • Are breastfeeding or are planning to breastfeed
  • Have a history of lung or breathing problems

Rybrevant generic

Rybrevant is the trademark brand name for amivantamab-vmjw manufactured by Janssen Pharmaceutical Companies of Johnson & Johnson. A generic version of amivantamab-vmjw is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Rybrevant savings through NiceRx.
