What is Scenesse?

Scenesse (afamelanotide implant) is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Clinuvel Inc. It is a melanocortin 1 receptor (MC1-R) agonist used to increase pain free light exposure in adult patients with a history of phototoxic reactions from erythropoietic protoporphyria (EPP).

EPP is a rare disease that causes intolerance to light (photosensitivity). It is caused by mutations leading to impairment in ferrochelatase (FECH) activity, an enzyme involved in heme production.

Phototoxicity is one of the more common symptoms of EPP, usually starting in early childhood as an intolerance to sun exposure, with people experiencing severe burning pain most often on the face and hands under direct sunlight and some artificial light. EPP is the most common porphyria in children and the third most common in adults.

What doses of Scenesse are available?

Scenesse is available as a subcutaneous implant, in the following doses: 16 mg.

Please read the medication labeling and always speak with a healthcare professional for medical advice about any changes to your dose so they can monitor and evaluate your condition.

Scenesse active ingredients

The Scenesse implant contains afamelanotide as the active ingredient. It increases the levels of eumelanin in the skin, helping to shield against UV radiation and visible light, including direct sunlight.

EPP can affect your quality of life, and lifestyle adjustments are required in order to avoid painful reactions. Sun protective clothing, tanning creams or sunscreens, and tinting of car and home windows can all be of some benefit.

Scenesse side effects

The most common side effects of Scenesse in clinical trials compared to placebo include:

  • Implant site reaction
  • Skin irritation
  • Nausea, dizziness
  • Respiratory tract infection
  • Oropharyngeal pain, cough
  • Fatigue, somnolence
  • Skin pigmentation, melanocytic nevus
  • Non-acute porphyria

In rare instances, Scenesse can cause more serious side effects. These can include:

  • Darkening of pre-existing nevi and ephelides. Regular full body skin examination is recommended every 6 months

Your doctor will assess the benefits of using Scenesse against your risk of side effects. Patients are encouraged to report negative side effects or adverse reactions of Scenesse to the FDA. Visit www.fda.gov/medwatch, or call 1-800-FDA-1088.
