What is Tavalisse?

Tavalisse is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Rigel Pharmaceuticals, Inc. It is a kinase inhibitor used for the treatment of thrombocytopenia in adult patients with chronic immune thrombocytopenia (chronic ITP) who have had an insufficient response to a previous treatment.

What doses of Tavalisse are available?

Tavalisse is available in tablet form, in the following doses: 100 mg, 150 mg.

Start Tavalisse dose at 100 mg orally twice daily with or without food. Increase to 150 mg twice daily, after 4 weeks if needed, to achieve a platelet count of at least 50 x 109 /L as necessary to reduce the risk of bleeding. Manage adverse reactions using reduction of dose, interruption of treatment, or stopping treatment. Discontinue Tavalisse after 12 weeks if the platelet count does not increase to a level sufficient to avoid clinically important bleeding.

Tavalisse active ingredients

Tavalisse contains the active ingredient fostamatinib disodium hexahydrate. Fostamatinib is a tyrosine kinase inhibitor shown to work against spleen tyrosine kinase (SYK). The major metabolite of fostamatinib, R406, prevents signal transduction of Fc-activating receptors and B-cell receptors. The fostamatinib metabolite R406 reduces antibody-mediated destruction of platelets.

Tavalisse side effects

The most common side effects of Tavalisse in clinical trials include:

  • Diarrhea, nausea, abdominal pain
  • Respiratory infection, chest pain
  • Dizziness
  • Increased ALT/AST
  • Rash
  • Fatigue

In rare instances, Tavalisse can cause more serious side effects. These can include:

  • Hypertension – monitor blood pressure every 2 weeks until stable
  • Hepatotoxicity -monitor with liver function tests
  • Diarrhea – if diarrhea becomes severe, interrupt, reduce or stop treatment
  • Neutropenia (decreased white blood cell count)

Tavalisse drug interactions

Tavalisse can interact with other medications. These include:

  • Strong CYP3A4 Inhibitors
  • Strong CYP3A4 Inducers
  • BCRP substrate drugs – rosuvastatin
  • P-Glycoprotein (P-gp) substrates – digoxin

This list is not exhaustive and other prescription drugs may interact with Tavalisse.

Tavalisse contraindications

You should not use Tavalisse if you:

  • Are allergic to the active ingredient fostamatinib
  • Are allergic to any of the other ingredients in Tavalisse
  • Are pregnant or are planning to become pregnant – Embryo-fetal toxicity. Tavalisse can cause fetal harm and should not be used in pregnant women. Females of reproductive potential to use effective contraception during treatment and for at least 1 month after the last dose
  • Are breastfeeding or are planning to breastfeed
  • Are younger than 18 years old

Talk to your doctor before using Tavalisse if you:

  • Are taking any of the medications that could interact with Tavalisse
  • Have high blood pressure
  • Have liver problems

Tavalisse generic

Tavalisse is the trademark brand name for fostamatinib manufactured by Rigel Pharmaceuticals, Inc. A generic version of fostamatinib is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Tavalisse savings through NiceRx.
