What is Tivdak?

Tivdak is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Seagen Inc., and Genmab US, Inc. Tivdak is a tissue factor-directed antibody-drug conjugate (ADC) and microtubule inhibitor used to treat adult cancer patients with recurrent or metastatic cervical cancer with disease progression on or after chemotherapy.

This use is approved under accelerated approval based on tumor response rate and durability of response. Continued approval for this indication may be based on verification and description of clinical benefit in confirmatory trials.

What doses of Tivdak are available?

Tivdak is available in injection form, in the following doses: 40 mg as a lyophilized cake or powder in a single-dose vial for reconstitution.

Tivdak is for intravenous infusion only. Not as an intravenous push or bolus. Tivdak is not to be mixed with or administered as an infusion with other medicinal products. The recommended dose is 2 mg/kg as an intravenous infusion over 30 minutes every 3 weeks until disease progression or unacceptable toxicity.

Tivdak active ingredients

Tivdak contains tisotumab vedotin-tftv as the active ingredient.

Tivdak side effects

The most common side effects of Tivdak in clinical trials include:

  • Hemoglobin decreased, lymphocytes decreased, leukocytes decreased, creatinine increased
  • Prothrombin international normalized ratio increased, activated partial thromboplastin time prolonged
  • Fatigue
  • Nausea
  • Alopecia
  • Epistaxis
  • Conjunctival adverse reactions (eye problems), dry eye
  • Diarrhea, and rash

In rare instances, Tivdak can cause more serious side effects. These can include:

  • Ocular toxicity – changes in the corneal epithelium and conjunctiva resulting in changes in vision, including severe vision loss, and corneal ulceration
  • Peripheral neuropathy
  • Hemorrhage
  • Severe, life-threatening, or fatal pneumonitis
  • Embryo-fetal toxicity

Tivdak drug interactions

Tivdak can interact with other medications. These include:

  • Strong CYP3A4 Inhibitors – may increase unconjugated monomethyl auristatin E (MMAE) exposure

Tivdak contraindications

You should not use Tivdak if you:

  • Are allergic to the active ingredient tisotumab vedotin-tftv
  • Are allergic to any of the other ingredients in Tivdak
  • Are pregnant or are planning to become pregnant – females of reproductive potential are advised to use effective contraception during treatment and for 2 months after the last dose. Male patients with female partners of reproductive potential are advised to use effective contraception during treatment and for 4 months after the last dose

Talk to your doctor before using Tivdak if you:

  • Are taking any of the medications that could interact with Tivdak
  • Have a moderate or severe hepatic impairment
  • Are breastfeeding or are planning to breastfeed

Tivdak generic

Tivdak is the trademark brand name for tisotumab vedotin-tftv manufactured by Seagen Inc. and Genmab US, Inc. A generic version of tisotumab vedotin-tftv is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Tivdak savings through NiceRx.
