What is Vyndaqel?

Vyndaqel is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Pfizer Inc. The FDA approval was based on data from the pivotal Phase 3 Transthyretin Amyloidosis Cardiomyopathy Clinical Trial (ATTR-ACT). The first global, double-blind, randomized, placebo-controlled clinical study to investigate a pharmacological therapy for the treatment of this disease. In ATTR-ACT, Vyndaqel significantly reduced the hierarchical combination of all-cause mortality and frequency of cardiovascular-related hospitalizations compared to placebo over a 30-month period. Also, individual components of the primary analysis showed a relative reduction in the risk of all-cause mortality and frequency of cardiovascular-related hospitalization.

Vyndaqel and Vyndamax are transthyretin stabilizers used for the treatment of the cardiomyopathy of wild type or hereditary transthyretin-mediated amyloidosis (ATTR-CM) in adults, to reduce cardiovascular mortality and cardiovascular-related hospitalization.

Transthyretin amyloid cardiomyopathy is a rare disease, where unstable transthyretin tetramers dissociate and result in a buildup of abnormal deposits of misfolded proteins that aggregate into amyloid fibrils and deposit in the heart. This causes the heart muscle to become stiff, eventually resulting in heart failure.

What doses of Vyndaqel are available?

Vyndaqel is available in capsule form, in the following doses: Tafamidis meglumine 20 mg and tafamidis 61 mg.

The recommended dose is either Vyndaqel 80 mg orally once daily, or Vyndamax 61 mg orally once daily.

Vyndaqel active ingredients

Vyndaqel contains the active ingredient tafamidis, a selective stabilizer of transthyretin (TTR). Tafamidis binds to TTR at the thyroxine-binding site, stabilizing the tetramer.

Vyndaqel side effects

The most common side effects of Vyndaqel in clinical trials include:

  • Cough, difficulty breathing, problems swallowing
  • Faster heart rate, dizziness
  • Hive, itching, swelling of eyes, face, lips, or tongue
  • Fatigue
  • Abdominal pain, diarrhea

Vyndaqel contraindications

You should not use Vyndaqel if you:

  • Are allergic to the active ingredient tafamidis meglumine
  • Are allergic to any of the other ingredients in Vyndaqel
  • Are pregnant or are planning to become pregnant – tafamidis may cause fetal harm when given to a pregnant woman
  • Are breastfeeding or are planning to breastfeed

Vyndaqel is the trademark brand name for tafamidis meglumine manufactured by Pfizer Inc. A generic version of tafamidis meglumine is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Vyndaqel savings through NiceRx.
