What is Xyntha?

Xyntha is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Wyeth Pharmaceuticals LLC, A subsidiary of Pfizer Inc. Xyntha works by temporarily raising levels of factor VIII in the blood to aid in clotting.

Xyntha, antihemophilic factor (recombinant), is indicated for use in adults and children with hemophilia A (congenital factor VIII deficiency) for:

  • On-demand treatment and control of bleeding episodes
  • Perioperative management
  • Routine prophylaxis to reduce the frequency of bleeding episodes

Xyntha does not contain von Willebrand factor, and therefore is not indicated in patients with von Willebrand’s disease.

What doses of Xyntha are available?

Xyntha is available as a lyophilized powder in single-use vials containing nominally 250, 500, 1000, or 2000 IU.

Xyntha is also supplied in kits that have both the powder and the diluent within single-use prefilled dual-chamber syringes, called Xyntha Solofuse.

The powder must be mixed with the diluent before injecting. If you store your medicine in the refrigerator, take the medicine and diluent vial out of the refrigerator and allow each to reach room temperature before mixing. After mixing, keep the mixture at room temperature and use it within 3 hours. If you have not used it in 3 hours, throw it away. Do not put mixed medicine into a refrigerator.

Xyntha is for intravenous use after reconstitution only. The required dose is determined by using body weight. For routine prophylaxis in adults and adolescents (≥12 years) the recommended starting regimen is 30 IU/kg 3 times weekly. The recommended starting regimen for children (<12 years) is 25 IU/kg every other day. Adjust the dosing regimen based on clinical response.

Please see full the full prescribing Information and always speak with a healthcare professional about any changes to your dose so they can monitor and evaluate your condition.

Xyntha active ingredients

The active ingredient in Xyntha, Antihemophilic Factor (Recombinant), is a recombinant antihemophilic factor, also called coagulation factor VIII. It is produced by recombinant DNA technology.

Xyntha side effects

The most common side effects of Xyntha in clinical trials include:

  • In adult and pediatric previously treated patients (PTPs) – headache, arthralgia, pyrexia, and cough

In rare instances, Xyntha can cause more serious adverse effects. These can include:

  • Anaphylaxis and severe hypersensitivity reactions
  • Development of neutralizing antibodies – If expected plasma factor VIII activity levels are not attained, or if bleeding is not controlled with an appropriate dose, perform an assay that measures factor VIII inhibitor concentration

Your doctor will assess the benefits of using Xyntha against your risk of side effects. Patients are encouraged to report negative adverse events or adverse reactions of Xyntha to the FDA. Visit www.fda.gov/medwatch, or call 1-800-FDA-1088.
