What is Zynlonta?

Zynlonta is an FDA (U.S. Food and Drug Administration) approved medication manufactured by ADC Therapeutics SA. Zynlonta is a CD19-directed antibody-drug conjugate (ADC), indicated for the treatment of adult patients with relapsed or refractory large B-cell lymphoma after two or more lines of systemic therapy, including diffuse large B-cell lymphoma (DLBCL) not otherwise specified, DLBCL arising from low-grade lymphoma, and high-grade B-cell lymphoma. This indication is approved under accelerated approval based on the overall response rate.

What doses of Zynlonta are available?

Zynlonta is available in injection form, in the following doses: 10 mg of loncastuximab tesirine-lpyl as a lyophilized powder in a single-dose vial for reconstitution and further dilution.

Zynlonta is given as an intravenous infusion over 30 minutes on Day 1 of each cycle, every 3 weeks. The recommended dosage is 0.15 mg/kg every 3 weeks for 2 cycles. 0.075 mg/kg every 3 weeks for the following cycles.

Premedicate with dexamethasone 4 mg orally, or intravenously twice daily for 3 days, starting the day before Zynlonta treatment starts. See the full prescribing information for instructions on preparation and administration.

Zynlonta active ingredients

Zynlonta contains the active ingredient loncastuximab tesirine-lpyl.

Zynlonta side effects

The most common side effects of Zynlonta in clinical trials include:

  • Thrombocytopenia (reduced platelets)
  • Increased gamma-glutamyltransferase
  • Neutropenia, anemia, hyperglycemia
  • Transaminase elevation
  • Hypoalbuminemia
  • Fatigue
  • Rash, edema, nausea
  • Musculoskeletal pain

In rare instances, Zynlonta can cause more serious adverse reactions. These can include:

  • Pleural effusion, pericardial effusion, ascites, peripheral edema, and general edema
  • Myelosuppression
  • Infections
  • Cutaneous reactions
  • Embryo-fetal toxicity

Zynlonta drug interactions

There is currently no data for Zynlonta interactions with other medications. However, other drugs may affect loncastuximab tesirine-lpyl, including prescription and over-the-counter medicines, vitamins, and herbal products.

Zynlonta contraindications

You should not use Zynlonta if you:

  • Are allergic to the active ingredient loncastuximab tesirine-lpyl
  • Are allergic to any of the other ingredients in Zynlonta
  • Are pregnant or are planning to become pregnant – use effective birth control during treatment and for 9 months after the last dose
  • Are breastfeeding or are planning to breastfeed

Talk to your doctor before using Zynlonta if you:

  • Are taking any of the medications that could interact with Zynlonta

Zynlonta generic

Zynlonta is the trademark brand name for loncastuximab tesirine-lpyl manufactured by ADC Therapeutics SA. A generic version of loncastuximab tesirine-lpyl is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Zynlonta savings through NiceRx.
