What is Ayvakit?

Ayvakit is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Blueprint Medicines Corporation. Ayvakit is a tyrosine kinase inhibitor used to treat:

  • Adult patients with unresectable or metastatic gastrointestinal stromal tumor (GIST) harboring a platelet-derived growth factor receptor alpha (PDGFRA) exon 18 mutation, including PDGFRA D842V mutations
  • Advanced systemic mastocytosis (AdvSM), including aggressive systemic mastocytosis (ASM), systemic mastocytosis with an associated hematological neoplasm (SM-AHN), and mast cell leukemia (MCL)

AdvSM is a rare hematologic disorder caused in most cases by the KIT D816V mutation. Ayvakit is not recommended for the treatment of AdvSM in people with low platelet counts.

What doses of Ayvakit are available?

Ayvakit is available in tablet form, in the following doses: 100 mg, 200 mg, and 300 mg.

Patients are selected for treatment based on the presence of a PDGFRA exon 18 mutations. The recommended dose is 300 mg orally once daily on an empty stomach. Take Ayvakit at least one hour before and two hours after a meal.

Ayvakit active ingredients

Ayvakit contains the active ingredient avapritinib, a targeted therapy.

Ayvakit side effects

The most common adverse reactions of Ayvakit in clinical trials include:

  • Edema
  • Nausea, vomiting
  • Tiredness, asthenia
  • Decreased appetite, diarrhea, constipation, abdominal pain
  • Changes in hair color
  • Increased lacrimation
  • Rash

In rare instances, Ayvakit can cause more serious side effects. These can include:

  • Intracranial hemorrhage
  • Central nervous system effects – cognitive impairment, dizziness, sleep disorders, mood disorders, speech disorders, and hallucinations
  • Embryo-fetal toxicity
  • Severe hepatic impairment

Ayvakit drug interactions

Ayvakit can interact with other medications. These include:

  • Strong and moderate CYP3A inhibitors – consider a reduction of the dose of Ayvakit if coadministration can not be avoided
  • Strong and moderate CYP3A inducers

This list is not exhaustive and other prescription drugs may interact with Ayvakit. Tell your healthcare provider about all the medicines you take, including prescription drugs and over-the-counter medicines, vitamins, and herbal supplements.

Ayvakit contraindications

You should not use Ayvakit if you:

  • Are allergic to the active ingredient avapritinib
  • Are allergic to any of the other ingredients in Ayvakit
  • Are breastfeeding or are planning to breastfeed

Talk to your doctor before using Ayvakit if you:

  • Are taking any of the medications that could interact with Ayvakit
  • Are a pregnant woman or are planning to become pregnant – Females and males of reproductive potential should use effective contraception during and for 6 weeks after the final dose
  • Are breastfeeding or are planning to breastfeed

Ayvakit generic

Ayvakit is the trademark brand name for avapritinib manufactured by Blueprint Medicines Corporation. A generic version of avapritinib is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Ayvakit savings through NiceRx.
