What is Prolastin?

Prolastin-C Liquid is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Grifols Therapeutics, Inc.

It is an Alpha1-Proteinase Inhibitor (Human) (Alpha1-PI) used for chronic augmentation and maintenance therapy in adults with clinical evidence of emphysema due to severe hereditary deficiency of Alpha1-PI (alpha1-antitrypsin deficiency). AAT deficiency is the number 1 known genetic risk factor for COPD and may be a contributing cause for up to 3% of COPD (chronic obstructive pulmonary disease) cases in the United States.

The effect of augmentation therapy with any Alpha1-PI on pulmonary exacerbations and on the progression of emphysema in Alpha1-PI deficiency has not been conclusively demonstrated in randomized, controlled clinical trials. Prolastin-C Liquid is not recommended for lung disease in patients in whom severe Alpha1-PI deficiency has not been found.

What doses of Prolastin are available?

Prolastin is available in injection form, in the following doses: approximately 1,000 mg in a single-use vial containing 20 mL of solution for injection.

The recommended dose of Prolastin-C Liquid is 60 mg/kg body weight, given intravenously once per week.

Prolastin is a liquid version of alpha 1-proteinase inhibitor that is not mixed with a diluent. Store Aralast, Prolastin, or Zemaira at cool room temperature, away from moisture and heat and use within 3 hours after mixing.

Store Prolastin in the refrigerator. Take the medicine out of the refrigerator and let it reach room temperature before preparing your dose. Once Prolastin has reached room temperature, you must use it within 1 month. Do not return it to a refrigerator.

Read the prescribing information and all medication guides or instruction sheets. Use the medicine exactly as directed. Always speak with your healthcare provider about any changes to your dose so they can monitor and evaluate your condition.

Prolastin active ingredients

Prolastin contains the active ingredient Alpha-1-proteinase inhibitor. It is used to replace an enzyme in patients with lung problems caused by low levels of alpha-1 antitrypsin.

Prolastin side effects

The most common side effects of Prolastin in clinical trials include:

  • Diarrhea
  • Tiredness, weakness
  • Symptoms of a cold
  • Headache, joint pain
  • Nausea
  • Bruising or bleeding where the injection was given

In rare instances, Prolastin can cause more serious adverse effects. These can include:

  • Difficulty breathing, chest pain
  • Dizziness
  • Seizures
  • Sudden numbness or weakness, problems with vision or balance, slurred speech
  • Severe hypersensitivity – stop Prolastin and initiate emergency treatment if hypersensitivity reactions occur

Because Prolastin is made from human plasma there is a very small chance it may carry infectious agents and you could get infections (including viral infections such as hepatitis, parvovirus B19, CJD), even though careful screening of donors, special manufacturing processes, and many tests are used to reduce this risk.

Prolastin drug interactions

Currently, no interactions are listed with other medications. Tell your healthcare provider about all other medicines you use, including prescription drugs and over-the-counter medicines, vitamins, herbal products, and supplements.

Prolastin contraindications

You should not use Prolastin if you:

  • Have a history of anaphylaxis or other severe systemic reaction to Alpha-1-proteinase inhibitor
  • Are allergic to any of the other ingredients in Prolastin
  • Have an IgA (immunoglobulin A) deficiency or antibody against IgA

Talk to your doctor before using Prolastin if you:

  • Are taking any of the medications that could interact with Prolastin
  • Are pregnant or are planning to become pregnant
  • Are breastfeeding or are planning to breastfeed

Prolastin generic

Prolastin is the trademark brand name for Alpha-1-proteinase inhibitor manufactured by Grifols Therapeutics, Inc. A generic version of the Alpha-1-proteinase inhibitor is not available. Generic drugs are generally cheaper than brand-name drugs, but you can still find Prolastin savings through NiceRx.
