What is Vyondys 53?

Sarepta Therapeutics Inc. has been granted FDA approval for Vyondys 53 (U.S. Food and Drug Administration) injection for the treatment of Duchenne muscular dystrophy (DMD).

Duchenne is caused by a mutation in the dystrophin gene, with one or more exons missing. This causes problems in the instructions for the making of dystrophin, and because of this, the body is unable to produce enough working dystrophin protein. Patients who receive treatment with Vyondys 53 must have a genetic test showing a mutation in the dystrophin gene that can be treated by skipping exon 53. Vyondys 53 is an exon-skipping therapy that allows the body to make a shorter form of the dystrophin protein, thereby raising dystrophin levels in the body.

Vyondys 53 is used for the treatment of DMD in patients who have a confirmed mutation of the DMD gene that is responsive to exon 53 skipping. It is approved under accelerated approval based on elevations in dystrophin production in skeletal muscle. Continued approval for this use may depend on the verification of a clinical benefit in confirmatory trials.

What doses of Vyondys 53 are available?

Vyondys 53 is available in injection form, in the following doses: 100 mg/2 mL in a single-dose vial.

Measure the patient’s glomerular filtration rate before commencing treatment, and administer as an intravenous infusion over 35 to 60 minutes.
